And now for some holiday fun… Our colleagues recently hosted Refeyn’s 4th employee-led blind taste test extravaganza at our Oxford HQ – Christmas Feastathon 2024. 🎄 The goal? To discover which Oxford supplier offers the best holiday-themed sandwich and best mince pie (a traditional British holiday dessert consisting of dried fruits and spices in a pastry shell). Participants rigorously sampled treats from Taylors Of Oxford, Pret, Marks & Spencer, Sainsbury's and Tesco. They were rated across five categories including: Taste (Fa la la la Flavor), Jingle Smell, and Appearance (Frost impressions). 🏆 Taylors emerged as the People's Champion, winning hearts with their seasonal delights. In a surprising upset, Marks & Spencer came in last place. Shout outs to our colleagues who made it all happen: R&D Engineer Charles Wright and R&D Manager Lee Priest, and to ‘tasting veteran’ and Senior R&D Engineer Nathan Curry, as well as to everyone who took part! 😋 What do you think we should test next time?

Refeyn, the pioneer of mass photometry, is delivering a disruptive new generation of analytical instruments that let you weigh individual molecules with light. Mass photometry offers new possibilities to characterise the function, structural composition and dynamics of biomolecules. Refeyn instruments measure the mass of individual molecules directly in solution, rapidly, and simply, revealing the true behaviour of molecules in near-native environments, providing scientists with key information about their mass distribution. Refeyn was spun out of Oxford University in 2018 to make mass photometry available globally. See how #MassPhotometry can be used for epitope binning in #antibody characterization in a recent paper from Ben Spiller's lab at Vanderbilt University School of Medicine. The authors created a library of #nanobodies against Streptococcus pyogenes Cas9 (SpCas9). After screening, three nanobody variants were found that tightly bound different epitopes in SpCas9 without interfering with its enzymatic activity or each other – a helpful toolbox for attaching genome-modifying enzymes or fluorescent tags to Cas9. Delighted to finally get this installed! New TwoMP #massphotometer from Refeyn, with a shiny #MassFluidix unit for low affinity interactions (apparently this is only the 2nd one of these in the UK!) The best thing is, this is core equipment and open for all users from across #Sheffield (and further afield) . With #MassPhotometry a simple dilution step and a few minutes is enough to analyze how different conditions affect protein behavior – like pH conditions in malaria parasite exonemes. This paper conclusively shows that the aspartic protease PMX is required for the maturation of the SUB1 proenzyme – which is in turn required to egress from red blood cells by Plasmodium falciparum. Chrislaine Withers-Martinez and colleagues from The Francis Crick Institute also provide structural details on how PMX cleaves the SUB1 prodomain and use mass photometry to describe the oligomerization behavior of SUB1 at the pH values found in P. falciparum exonemes. We teamed up with Cube Biotech, whose copolymer-derived native nanodiscs make it possible to solubilize and stabilize membrane proteins in one step, without detergents🧬. In just minutes, mass photometry gave clear insights into sample purity and heterogeneity, and protein interactions of G-protein coupled receptors (GPCRs) within copolymer nanodiscs 🔬💡. The results confirm that mass photometry is an excellent tool for quality checks before diving deeper into structural and functional studies.
