🤦♀️ Many traps are possible with solid state ! That is why a complete knowledge of cristalline forms is very important for pharma development. ❕ The more we know about polymorphic forms, the better. If having fully characterized the targeted crystal form is necessary, knowledge on the other undesired solid forms is critical and highly recommended. Why ? Because knowing in which conditions the undesired crystal forms could appear gives us the opportunity to control them and thus to be sure that they will not arrive, not disturb the pharma development. Which traps are we talking about ? Hydrate/solvate, a more stable polymorphic form, monotropy/enantiotropy, non congruente solubility, ... 👉 Solid state screenings allow to generate such data. 👨🔬 Holodiag is your expert CRO for polymorphs, cocrystals and salts screenings. #screenings #crystals #pharma

New Article on Deformulating LNPs Camille Malburet, Stéphanie Prizzon and co-workers from Sanofi recently published a valuable paper about deformulating #LNPs before #mRNA integrity analysis. They compared spin column extraction protocols and isopropanol ammonium acetate precipitation. Spin column extraction with silica media is promising as all biologists and biochemists know from historical total #RNA/DNA extraction approaches. However, many of these tried and true protocols are still using chloroform and phenol. The Sanofi team wrote a sentence wondering if non-toxic solvents could be used for silica adsorption. I wonder too! Deformulated mRNA are tested for integrity and purity by CE in this work, but it is reasonable to propose that an effective deformulation procedure could also be paired with size exclusion #HPLC chromatography. 📚Article: https://lnkd.in/ezJ-rMzu

What's driving Medicinal Chemists to go with the goldilocks is the ability of Macrocyclics to provide functional diversity and stereochemical complexity within partially rigid structures, while offering new chemical spaces and novel intellectual property to play with. A growing numerous applications of macrocyclizations in lead optimization, to demonstrate that despite their large size, they tends to possess adequate cell permeability and bioavailability, which enables them to reach their intracellular targets after oral administration. An excellent review providing a detailed case study of the application of macrocyclization over the past 5 years, starting from the initial analysis of acyclic active compounds to optimization of the resulting macrocycles for improved efficacy and drug-like properties. https://lnkd.in/gPj55D23

The Vapourtec Peptide-ExplorerLT is designed for scientists who need to explore and optimise the synthesis of linear peptides. The system produces the same high-quality peptides as Vapourtec's existing Peptide Explorer, but has a more compact footprint and is available at a lower price. CSO Manuel Nuño explained: "Peptide chemists can now access all the benefits of Fast Flow Phase Peptide Synthesis (FF-SPPS) at a price that is comparable with traditional, automated batch synthesizers" Click here to find out more https://lnkd.in/eqviCqqK #PeptideSynthesis #Vapourtec #PeptideChemists

DMPE (14:0 PE), or 1,2-Dimyristoyl-sn-glycero-3-phosphoethanolamine, is a phospholipid. This compound is made up of a glycerol backbone, with two myristoyl fatty acid chains attached. The "14:0" designation denotes that the fatty acid chains are myristoyl, meaning they consist of 14 carbon atoms with no double bonds. This specific phospholipid is of interest in biological and chemical research due to its role in cellular membranes and its potential applications in encapsulation and drug delivery systems. #DMPE #Dimyristoyl #encapsulation #membranes #drug #myristoyl #fatty #acid #chains https://lnkd.in/gKnuc_Sd
