Optimizing Cell Density: A Key to Robust Cell-Based Assays Proliferation and Viability As you explore cell densities for assay development, tracking cell proliferation and viability yields valuable insights. Cell viability assessments utilizing indicators like ATP levels or dyes that distinguish between live and dead cells offer insights into density-related phenomena, such as sparse or overcrowded conditions that can prompt cellular demise. Among the frequently employed dyes are Calcein-AM, utilized for live cell detection, and propidium iodide, ethidium homodimer, or trypan blue, employed for identifying dead cells. Proliferation assays such as BrdU or EdU incorporation can provide quantitative measures of expected cell division activity. This article shows that the G-Rex method reduces the residual TCR/CD3+ contaminations 45x more than their previous method while expanding the target CAR T cells, yielding in an ultrapure product of 99.99 % The new depletion method used focuses on coculturing target CAR T cells with an NK-92 cell line in G-Rex to purify their CAR T cells and promote CAR T cell expansion at the same time.
