Here Anders et al describes the effect of lipid nanoparticle encapsulation on ribonucleic acid via ion mobility mass spectrometry 🛎 Overview: Studying the contents of LNPs, such as RNA, can be difficult because most research focuses on the overall LNP structure. Many questions remain about how RNA interacts with LNP lipids and water, causes LNP instability, and forms complex structures. Understanding RNA in LNPs can improve RNA-based therapies. 🎯 Summary: Anders et al study employed IM-MS and CIU to analyze RNA cargo released from LNPs in the gas phase. Anders et al assessed WT mt-tRNA Leu (UUR) in LNPs made with various lipids, finding differences in RNA structure and stability. The presence of Mg2+ also impacted these differences. Multiple factors influence additive or synergistic effects of PFAS mixtures on rainbow trout gill cells A new study used the immortalized rainbow trout gill cell line (RTgill-W1) to investigate cell viability and phospholipidosis after exposure to different PFAS compounds. Most compound mixtures showed synergistic effects on cytotoxicity that varied with dose and ratio. For example, PFOS and PFECHS were additive at low doses but synergistic at high doses. CAIN - A CRISPR tool that "overrides Mendelian inheritance" to enable 99% gene transmission 🧬Darwinian natural selection directs gene inheritance by Mendel's laws, where an equal chance is typical for alleles to pass on to the next generation. 🧬Artificial selection (and sometimes natural selection) enables a "super-Mendelian inheritance" where certain genes are inherited at rates greater than the expected 50%, potentially allowing these genes to spread through populations even if they are detrimental to organisms. Could we use this mechanism to manipulate natural populations by introducing alleles that benefit us even if they harm some plant species, or to eliminate species that are not useful in our fields? 🧬Researchers now developed a plant gene drive system called "CRISPR-Assisted Inheritance utilizing NPG1 (CAIN)", which uses a toxin-antidote mechanism in the male germline to override Mendelian inheritance in plants. 🧬CAIN uses a CRISPR-Cas9 construct that disrupts the key gene responsible for pollen germination (NPG1), which is the "toxin" part of the tool . The "antidote" is a recoded, CRISPR-resistant NPG1 gene that rescues functionality, but only in pollen cells containing the gene drive. 🧬In the proof of concept study, researchers were careful to prevent unintended spread of this mechanism to natural populations. Using a Vertical-Wheel Bioreactor to produce extracellular vesicles from human forebrain spheroids: in their latest work, Chang Liu, Yan Li at Florida State University and collaborators differentiated human induced pluripotent stem cells into forebrain spheroids using Vertical-Wheel Bioreactors. Nice example of how loop residues can affect the binding affinity of G4 binding proteins. Here the Kd for an engineered DARPin for a G-quadruplex is increased 100 fold by inserting additional thymines in a loop.
