  • [New study] Lentiviral vectors are being increasingly used as tools for stable integration of large gene inserts into the genomes of both dividing and non-dividing cells. 𝐑𝐞𝐜𝐞𝐧𝐭 𝐚𝐧𝐢𝐦𝐚𝐥 𝐬𝐭𝐮𝐝𝐢𝐞𝐬 𝐡𝐚𝐯𝐞 𝐬𝐮𝐜𝐜𝐞𝐬𝐬𝐟𝐮𝐥𝐥𝐲 𝐝𝐞𝐦𝐨𝐧𝐬𝐭𝐫𝐚𝐭𝐞𝐝 𝐭𝐡𝐞𝐢𝐫 𝐮𝐬𝐞 𝐢𝐧-𝐯𝐢𝐯𝐨 To enable more widespread use, as well as development of in-vivo therapies in humans, better downstream processes for lentiviral purification are needed. In this work, we present the results of lentiviral downstream optimisation on CIM® QA monolith with the use of DoE. 💡DoE enabled an increase in infectious lentiviral recovery to 70%.   • [The world of mRNA] Learn how we’ve shortened the process of production and purification of tRNA from approximately a day and a half to 6 hours! ⏱️ 💡 Our study also demonstrated how at-line HPLC monitoring can be used to monitor, and improve, production yields of IVT-derived tRNAs. 𝐅𝐫𝐨𝐦 𝐦𝐑𝐍𝐀 𝐭𝐨 𝐭𝐑𝐍𝐀 Though the IVT monitoring approach is now widely used for mRNA applications, we show that adjustments in HPLC peak interpretation are required to apply the methodology to tRNA.   • [Improved performance] Chromatographic purification of pDNA is often a bottleneck due to low dynamic binding capacities (DBC), pDNA recovery and/or low selectivity of columns. 𝐇𝐨𝐰 𝐜𝐚𝐧 𝐰𝐞 𝐢𝐧𝐜𝐫𝐞𝐚𝐬𝐞 𝐃𝐁𝐂? 🔎 The goal of our investigation was to increase the DBC for pDNA on a monolith stationary phase by grafting the surface with linear polymethacrylate brushes, while retaining high pDNA recovery and chromatographic selectivity between pDNA and RNA impurities. 𝐔𝐩 𝐭𝐨 8-𝐭𝐢𝐦𝐞𝐬 𝐥𝐚𝐫𝐠𝐞𝐫 𝐃𝐁𝐂 Dynamic binding capacities for 7.3 kpb pDNA were up to 8-times larger compared to non-grafted CIMmultus 1 ml columns.   • [New study] By modifying pH, NaCl, MgCl2 and Arginine concentrations, using 96-well format and DoE, purification conditions can be optimised. 🔎 𝗟𝗲𝗻𝘁𝗶𝘃𝗶𝗿𝗮𝗹 𝗱𝗼𝘄𝗻𝘀𝘁𝗿𝗲𝗮𝗺 𝗽𝗿𝗼𝗰𝗲𝘀𝘀 𝗼𝗽𝘁𝗶𝗺𝗶𝘀𝗮𝘁𝗶𝗼𝗻 Currently, lentiviral based therapies are primarily ex-vivo, such as CAR- T, however recent animal studies have successfully demonstrated their use in-vivo as well. 💡 In this study, we present the results of lentiviral downstream optimisation on CIM® QA monolith with the use of DoE. 𝗧𝗵𝗲 𝗿𝗲𝘀𝘂𝗹𝘁? Optimised conditions from QA monolithic 96-well plates were successfully transferred to CIMmultus QA 1 mL Monolithic Column, where 70% average infectious recovery was achieved.
