  • The occurrence and dietary intake related to the presence of microplastics in drinking water in Saudi Arabia

    Environmental Monitoring and Assessment

The implications and health effects of microplastics (MPs) ingestion are still unclear, yet researchers and organizations around the world are increasingly examining the levels of microplastics and nanoplastics in the environment. This study investigated the presence and the quantity of microplastics in bottled and tap water from five regions in Saudi Arabia and estimated the dietary intake of microplastics. Thirty samples of drinking water were collected from the retail markets in Saudi Arabia. The samples included plastic bottled drinking water, 2 glass bottles, and 2 samples of tap water to represent ground water and desalinated seawater. Sample preparation in the laboratory involved a vacuum-assisted filtration with an inorganic filter membrane (0.2-µm pore size). Identification and classification of microplastics particles using Fourier-transform infrared microspectroscopy (FTIR microspectroscopy). The particle size …

  • Mapping QTL underlying fruit quality traits in an F1 strawberry population

Journal of horticultural science and biotechnology

Understanding how strawberry (Fragaria x ananassa) nutritional and quality traits are genetically regulated and correlated is an essential step towards improving marker-assisted breeding programmes in this crop. A first step to achieve this goal was to construct a single nucleotide polymorphism (SNP)-based genetic map of 140 F1 individuals of a cross between the 2 octoploid parents ‘Redgauntlet’ and ‘Hapil’. The map consisted of 3933 SNPs distributed over 28 linkage groups, representing the 7…

The map consisted of 3933 SNPs distributed over 28 linkage groups, representing the 7 homoeologous groups expected in Fragaria (2n = 8x = 56), and covered a total length of 2624.7 cM with an average resolution of 0.7 cM/SNP. Two overlapping subsets of F1 individuals were evaluated in the field and glasshouse at East Malling Research and the University of Reading, respectively. Negative correlation was observed between Pelargonidin and the three traits L*, a*, and b*, whereas positive correlations were observed within L*, a*, and b*, and between Ellagic acid, Pelargonidin, and Cyanidin at both trial locations. Quantitative trait loci (QTL) mapping revealed 29 significant QTL associated with the measured traits and mapped over 16 linkage groups. Two QTL for fruit fresh weight were co-located between day 1 and day 4 of shelf life, accounting for over 62% of the variation. These data will enhance our understanding of the genetic basis and correlation of strawberry quality traits and provide the basis for refining QTL that underpin the genetic regulation of these quality traits.

  • Characterization of a new octoploid strawberry breeding population

University of Reading - PhD Thesis

Among the Fragaria species, the cultivated strawberry Fragaria × ananassa (2n=8x=56) is the most economically valuable crops. After harvest, they are extremely perishable, have a short shelf life, and are susceptible to mechanical injuries and physiological decay due to loss of tissue integrity, sensitivity to fungal diseases, and have a large surface area, which lacks an outer protective rind. Therefore, maintaining high nutritional values in the berry fruit whilst maintaining high fruit…

Therefore, maintaining high nutritional values in the berry fruit whilst maintaining high fruit quality requires an understanding of the genetic and environmental effects on each trait, and how different traits are associated with each other. Mapping traits on the linkage map using a Quantitative Trait Loci (QTL) approach is the first step to identify the underlying gene(s) and to explore their effects and interactions. It will improve our understanding of the genetic control of measured traits and facilitates molecular marker development. Therefore, it can be used to improve plant-breeding efficiency at the molecular level, which significantly reduces the breeding time and cost of phenotyping. The overall aim of this study was to characterise the variation in quality traits among the F1 mapping progeny derived from a cross of Redgauntlet x Hapil (RGxH) strawberry cultivars. These traits include total soluble solids (TSS), titratable acidity (TA), fresh weight, surface colour, firmness, and phenolic content. This thesis presents two areas of work. First, using a novel high-density single nucleotide polymorphism (SNP) linkage map, phenotyping plant characteristics of RG x H progeny enabled the detection of QTL linked to traits associated with shelf life (at 4 °C). Subsequent QTL …
