On October 22, 2024, a research team from the Van Andel Institute published a study in #Cell Chemical Biology on lipid #metabolism and #ferroptosis, bringing new insights to the field of #ferroptosis research. The study found that although limiting extracellular lipids reduces the levels of polyunsaturated fatty acids (#PUFAs) within cells, lipid-deprived cancer cells exhibit increased sensitivity to ferroptosis. Under lipid-restricted conditions, cancer cells stimulate the transport of PUFAs, which are released from triglycerides and converted into highly unsaturated PUFAs, such as arachidonic acid and adrenic acid. These PUFAs then accumulate in phospholipids, thereby enhancing sensitivity to ferroptosis. We aimed to study their interactions with G-quadruplex (G4) structures as potent G4 ligands and telomerase inhibitors. We used a PCR-amplified guanine-rich region from the human beta-globin gene, oligonucleotide from human telomeres (a-coreTT), an oncogene (c-kit), and SK-HEP-1 adenocarcinoma cells to characterize those compounds’ binding and stabilizing abilities to G4 structures and anti-cancer potential. All results obtained through UV-visible and fluorescence spectroscopies, agarose gel electrophoresis, and MTT assay on SK-HEP-1 adenocarcinoma cells were in good agreement. Compounds 4 and 6 are promising DNA-binding and cytotoxic compounds with a relatively antiproliferative effect on the selected tumour. In all studies, the formal positive charge carrier, compound 6, showed higher activity in terms of anti-cancer effects. However, there is still insufficient information about the features of the biosynthesis of these compounds and the conditions for their production. In this paper, the influence of an inorganic nitrogen source on the growth of a suspended culture of medicinal asparagus, the accumulation of phenolic compounds in the culture and culture liquid, and the synthesis of saponins is studied. A callus and suspension culture of medicinal asparagus Asparagus officinalis L. were obtained under in vitro conditions. For further research, concentrations of growth regulators 1 mg/L kinetin and 1.5 mg/L 2,4-D were selected. The fastest growth of the suspension culture was established on the medium with a concentration of potassium nitrate of 1900 mg/L and ammonium sulfate of 250 mg/L. Individual explants demonstrated spontaneous differentiation of callus tissue that affected the increase in callus mass and the ratio of the concentration of exophenols to the total mass of the explant. The highest concentrations of endo- and exophenols were obtained on a medium without ammonium nitrogen sources. A positive effect of the medium with a concentration of ammonium sulfate of 500 mg/L on the synthesis of the main steroidal saponins of Asparagus officinalis L. was determined. As a result of the study, a modified MS medium was developed that promotes the accumulation of steroid saponins in a suspension culture of medicinal asparagus. I'm pleased to announce that my recent research paper, titled "Differential Tissue Abundance of Membrane-Bound Drug-Metabolizing Enzymes and Transporter Proteins by Global Proteomics (https://lnkd.in/gZ8svtKv)," has been published in the Drug Metabolism and Disposition journal. We quantified the abundance and compositions of drug-metabolizing enzymes and transporters (DMETs) in pooled human liver, intestine, and kidney microsomes using an optimized novel sequence coverage-informed total protein approach (SC-TPA). Additionally, we identified high variability in the levels of intestinal DMEs (e.g., CYP3A4, CYP3A5, UGT2B17, CES2, and MGST2). The data generated regarding DMET abundance in the human liver, intestine, and kidney hold significant potential for informing the development of physiologically based pharmacokinetic (PBPK) models, particularly in explaining the role of enzymes and transporters in bioavailability and tissue-specific toxicity.
