In this month's #ComplementinLifeScience, a recent article reports the use of #riliprubart in treating cold agglutinin disease (#CAD). Riliprubart is a second-generation inhibitor that selectively inhibits the activated form of C1s. CAD is a rare autoimmune disease that causes hemolytic anemia mediated by the classical complement pathway (CP). In a phase 1b clinical trial, the safety and efficacy of riliprubart were assessed in CAD patients. Riliprubart's activity on CP and AP was determined by #Wieslabassays. The lack of change in Wieslab AP activity from baseline to the end of the study demonstrated its specificity for CP. The study also reports that in this cohort, a single dose led to a reduction in hemolytic complement, resolution of hemolysis, and correction of anemia, with responses maintained over the 15 weeks of the study with no serious adverse events.
