My son, Charles, has Phelan-McDermid syndrome (PMS), caused by the loss of the SHANK3 gene. In addition developmental delay and autism, he suffers from Lennox-Gastaut epilepsy. The silver lining of his terrible situation is that at least he has a genetic diagnosis, and that has enabled us to imagine a future where he has access to truly disease-modifying treatments.  At CureSHANK our mission is to accelerate treatments that can give Charles and others with PMS better quality of life. Collaboration between statistics and pharmacometrics has proceeded in fits and starts over the last three decades, but recent developments hold the promise of a more robust and fruitful dialogue. Among these developments are the anticipated release of ICH guideline M15 for model-informed drug development, the formal recognition of the "SxP SIG" as a scientific working group of the Biopharmaceutical Section of the ASA, and the increasing recognition within the statistical community of the value of causal inference based on non-randomized comparisons. This discussion will seek to brainstorm ways to capitalize on these and other developments in order to promote broader understanding of pharmacometric questions, perspectives, and methods within the statistical community.
