To mitigate some of the biggest risks in drug development, it’s essential to gather as much information about a candidate early on in development to make better, more informed decisions further down the line. This is where "developability" comes in, to understand whether the drug can be made and if it will work. At the recent Biologics US conference, our Sr. Director of Bioassays, Dr Erika Kovacs, presented a scientific poster entitled: "Rapid Developability Assessments to Assist Lead Candidate Selection." The poster highlights our robust developability platform, which integrates in silico approaches with characterization and stability studies to determine the selection of the best candidate to progress. Using a panel of 7 mAb sequences, the poster details the different computational tools employed to identify sequence liabilities and immunogenicity risks, while advanced biophysical techniques evaluated manufacturability and stability. The process showcased in this #biotechanimation is a novel patented cell-based assay for studying GPCR heteromers. Our client has developed a cell-based receptor-HIT assay designed to detect and study GPCR heteromers effectively. This platform is versatile, capable of working with various molecular targets, thereby minimizing the risk of unidentified signaling interactions. The patented assay functions as a three-component system, involving an unlabeled GPCR, a luciferase-fused second GPCR, and a fluorophore-fused signaling molecule. Ligand binding to the unlabeled GPCR and subsequent heteromerization with the labeled GPCR lead to signal generation due to proximity between the fluorophore and luciferase. This proximity-based mechanism ensures a robust assay with minimal background noise, especially when GPCRs form homodimers or in the absence of a ligand. The unique capabilities of this receptor-HIT assay make it ideal for repurposing approved compounds, identifying orphan receptors, and facilitating new drug discovery efforts.
