Meet WARIO, a novel approach to characterize ensembles of (highly-)flexible proteins. WARIO incorporates the probabilistic nature of #IDPs by capturing the ensemble information within a weighted family of contact maps. This method leverages a refined contact definition based on inter-residue geometry and sequence context. Subsequently, clustering these contact-based descriptors unveils representative structural features of the entire ensemble. Notably, WARIO effectively identifies even transiently populated structures. We demonstrate WARIO's superiority over existing methods in capturing crucial structural features of highly flexible proteins through several use-cases.
