Dr Cosmas Mafusire is a senior postdoctoral fellow in the Biophysics Group. He completed his PhD on laser beam propagation through random media in 2012. He is working on using bioinformatics and chem-informatics to study the role of various complex molecules such as CP24, CP26 and CP29 in photosynthetic systems. 'At the heart of photosynthesis is the remarkable efficiency of the energy-capturing phase, coupled with the ability of photosynthetic organisms to dissipate harmful excess energy through specialized mechanisms. Understanding these processes is crucial for designing efficient solar cells that mimic these natural conditions. My research focuses on investigating methods to accurately model natural systems by developing a quantum mechanical understanding of such processes. Central to our efforts is the calculation of an accurate Hamiltonian, as the success of the model hinges on accurately representing the Hamiltonian of natural systems.' The project is a collaboration between ILL - Institut Laue Langevin (Lionel Porcar), Federica Sebastiani and my team at Department of Pharmacy, University of Copenhagen. We aim at establishing neutron-based approaches for the analysis of the structural reorganization of a protein ensemble during the transition from the dilute to the dense phase of proteins and eventually to the solid state. FAPESP Postdoctoral Position on in situ Cryo-Electron Tomography With the Aquilos 2 cryo-FIB soon available at our Institute (https://lnkd.in/dcWt8QWd), allow me to briefly advertise a fellowship opportunity in my lab: This project to be developed at the São Carlos Institute of Physics (IFSC), University of São Paulo (USP) in Brazil, aims to study the in situ structure of mitochondrial filamentous enzymes involved in the metabolic adaptation of tumors and their effects on organelle ultrastructure and morphology upon filamentation. This research will be an extension of Adamoski et al., NSMB 2024 (doi: 10.1038/s41594-023-01118-0), focusing on more profound questions with cutting-edge methodologies, such as correlative light-electron microscopy (CLEM), cryo-electron tomography in lamella (cryo-ET) and expansion microscopy (ExM), that can also be applied to other proteins in the lab. Methodological Responsibilities: 1) Sample Preparation: Engage in cell culturing, vitrification, and lamellae preparation in our lab at the IFSC/USP; 2) Data Collection and Processing: Perform high-resolution tomographic data collection using top-tier microscopy facilities in Brazil and internationally. Data processing will be done in-house. Requirements: 1) PhD in Chemistry, Biochemistry, Molecular Biology, or related; 2) Experience in Cell Biology; proficiency in cryo-FIB and cryo-ET, or single-particle cryo-EM is a big plus; 3) Strong analytical, collaboration, and English language skills. Prof Tjaart Krüger is the leader of the Biophysics group. He was awarded his PhD cum laude at the Vrije Universiteit Amsterdam in 2011 and then completed a 20-month postdoctoral fellowship at the same university. He worked on single-molecule spectroscopy of the main light-harvesting complex of plants (LHCII) and extended the research to a few other plant antenna complexes during his postdoc. He has received numerous awards for his work, including a B2 rating from the NRF in 2024, the CSIR National Laser Centre Innovation Excellence Award in 2020, the Meiring Naudé Medal from the Royal Society of South Africa in 2019 and the University of Pretoria Exceptional Young Researcher’s Award in 2018. His work involves understanding the fundamental properties of the light-harvesting protein complexes of photosynthetic organisms, which are amazing molecular machines. They use quantum mechanics to optimise their functions, a property that has captivated physicists for the past few decades. They also feature as light-sensitive nano-switches to maintain a delicate balance between their light-harvesting and photoprotective functions. These systems are also a great source of inspiration for finding green, sustainable energy solutions for humankind. Some other applications of his work include quantum biotechnologies such as sensors, molecular switches, nanotechnology and biodegradable photoactive materials. Prof Krüger has chosen biophysics because it provides the opportunity to learn about living systems using physics: 'It's a great privilege to live in an era where technology combined with the principles of physics enables us to learn about life’s processes at an exceptionally deep level of detail – probably the most fundamental level of detail! There is so much to learn in this field that research will never get boring.' He also appreciates the potential of this field to address important global issues, and the opportunity it provides to do pioneering work in South Africa and on the African continent to develop this field of research. His talents are not limited to physics! He loves music, especially classical music. He plays piano, violin, organ and loves to compose music. He (unofficially) published his first book of compositions at the end of his Master's and since then he's been expanding the list of pieces. He says that 'hopefully I'll soon get time to round them off. The main problem is that when I start working on the previous compositions, I get ideas for new ones...
