𝗟𝗜𝗙𝗘 𝗢𝗙 𝗔 𝗕𝗜𝗢𝗖𝗛𝗘𝗠𝗜𝗦𝗧𝗥𝗬 𝗦𝗧𝗨𝗗𝗘𝗡𝗧 (𝗟𝗔𝗕𝗦) FINDING MY DREAM SEMINAR RESEARCH TOPIC. Finding a topic for my seminar work was really challenging. As a newbie without prior knowledge on scientific research, I was so lost in the sea of many options, suggestions and confusions. Some of my course mates were of the opinion that they won't take any seminar topic that would stress them, connsidering the fact that exam is just 4 weeks ahead, we just just had 3 weeks to conclude the research and defend it, plus exam preparation. Hence, they opted for cheap topics they can get by. I had my goal, I want to be a renown researcher, and as such, I had to take novel milestones to reach there, which starts from this undergraduate stage. I want to do something no one has ever done. I wanted to do a literature review that I'll be proud of and my supervisor will beat his chest and say "this is my student." These were my concerns and worries. I did a little oral research on the past research work senior colleagues did and I discovered that many did their work just to get the grade and pass. Only few impressed me. Reaching out to some of my course mates to discuss my concerns, some of them were like "chinenye, no do pass yourself oo." I knew I wanted to do a research on cancer, but cancer research is very vast that I was confused on which aspect to specialise. This initiative was birthed during my industrial training period at Center for Molecular Biology and Biotechnology, MOUAU. There I assisted a lab technician to conduct a research on the mutation of BRCA 1 & 2 genes - mutation in these genes is responsible for 80% of breast cancer. So assisting him in that research gave me an in-depth knowledge about cancer cells and the desire to procure solution to cancer. There, I told myself, "I'm going to do a research on cancer." Being an exceptional Biochemistry student is not easy, you'll have to work for it. Watch out for my post tomorrow on how I finally landed my dream seminar research topic. I am thrilled to announce the publication of my PhD research! 🎉📜 Our study delves into a fascinating question: How does a cell distinguish the same ligand acting on the same receptor but producing completely opposite signals and effects? 🤔🔍 To answer this, we used pituitary tumor cells, which cannot form tumors in mice due to their senescent features. Knocking out IL-6 in these cells transformed them into tumorigenic ones, shutting down senescence! Our key findings include: 🔬 Intracellular IL-6 Induced Senescence: Our question was whether IL-6 re-expression is enough to reestablish the senescence of these IL-6 knockout cells. As IL-6 is a cytokine that is secreted, we aimed to analyze the secretion-independent effects since its proliferative action through membrane receptors is well characterized. We observed that IL-6 induced senescence only when retained intracellularly, suggesting the senescent signal originates from within the cell. In human therapy-induced senescent cells, where IL-6 is relevant in tumorigenesis development, intracellular retention of IL-6 also showed pro-senescence effects. 🔬 Mechanistic Insights: We found that this differential senescent signaling involved cytosolic chromatin fragments and intracellularly expressed IL-6R. IL-6 acts as an amplifier of the senescent signal through an intracrine mechanism, relying on the cGAS-STING pathway.  Therefore, therapies that involve neutralization of IL-6R expressed on the cell membrane block IL-6's proliferative signal but might not affect this senescent intracrine action, which could represent a resistance mechanism in tumor biology. Thrilled to share my experience at the inaugural Federation of American Societies for Experimental Biology (FASEB) Hippo Pathway Conference! 🧬🔬 As a PhD student, I had the incredible opportunity to present my thesis work on therapeutic targeting of Hippo pathway in sarcomas. This conference was a pivotal moment in my scientific journey, bringing together leading researchers exploring the fundamental biology and therapeutic potential of the Hippo pathway. Key highlights: -Gained insights into Hippo signaling mechanisms and their critical role in cancer, stem cell development, and inflammation -Networked with experts developing innovative targeted therapies -Attended a fantastic Career Development Workshop that provided invaluable guidance for early-career researchers The most exciting aspect was the opportunity to learn about unpublished research and witness the collaborative spirit of the scientific community. The conference underscored the immense potential of Hippo pathway research in understanding disease mechanisms and developing breakthrough treatments. 🎓 PhD Defense Announcement 🎓 I am pleased to share that I successfully defended my PhD thesis on August 22, 2024, at the University of Namur, Belgium, where I earned my Doctorate in Biomedical and Pharmaceutical Sciences. My research focused on understanding a syndromic neurodevelopmental disorder (NDD) within a Lebanese family, linked to a homozygous stop-gain mutation (p.R151*) in the BOD1 gene. This mutation, associated with intellectual disability and other mild dysmorphic and endocrine symptoms, led us to explore its effects on protein expression and brain development. Using CRISPR-Cas9, we generated a knock-in mutation in HEK293T cells, revealing a truncated and unstable Bod1 protein, significantly impacting its function. Additionally, a Bod1 knock-out mouse model demonstrated notable cognitive deficits and structural brain changes, particularly in females, underscoring BOD1’s critical role in neurodevelopment. This research contributes valuable insights into the molecular mechanisms underlying BOD1-related syndromic intellectual disability and highlights the broader implications for neurodevelopmental disorders. I extend my heartfelt gratitude to my advisors, Pr. Olivier De Backer ,Pr. Eliane Chouery, as well as to the Professors and Jury members: Thierry Arnould, René Rezsohazy, Charles Nicaise, and Francoise Goflot. A boring Monday afternoon transformed into an exciting one, particularly for two individuals, as the Nobel Prize in Physiology and Medicine was announced on 7th October, 2024. Meet the latest Nobel Laureates, Victor Ambros and Gary Ruvkun! They received this prestigious award for discovering microRNA, a very small molecules found in the cells which play a role in regulating the functions of genes. Their discovery goes all the way back to 1993, when Victor’s lab first identified miRNA in a nematode called C. elegans. Apparently, at the same time, Gary Ruvkun discovered the target of miRNA. Victor Ambrose carried out his graduate research (Ph.D.) in genome structure at Massachusetts Institute of Technology (MIT) from 1976 to 1979. He then continued as a post-doctoral researcher in MIT, where he focussed on developmental timing in C. elegans. Presently, he is a professor at UMass Chan Medical School, USA. Gary Ruvkun began his academic journey in 1973 as a Biophysics undergraduate at the University of California, Berkeley. He went on to complete his Ph.D. in Biophysics from Harvard University in 1982. Currently, he is affiliated with the Harvard Medical School as a Professor of Genetics. “We weren’t thinking that this is going to win a Nobel Prize, we were thinking this is really interesting. As the field exploded, which is just a joy to watch, then there was a sense that this is the sort of field, the sort of sea-change that gets awards and things. But that took a long time and was an unbelievable pleasure to watch, to participate in” was what Dr. Ruvkun had to say during a telephonic interview. 🧬The mechanism of miRNA is relatively straightforward. Unlike double-stranded DNA, miRNA is a single-stranded molecule that targets messenger RNA (mRNA) of specific genes. When miRNAs bind to their target mRNAs, they facilitate gene silencing by preventing the mRNA from being translated into a functional protein. This mechanism serves to be crucial in normal developmental processes in animals and plants. miRNA are now being employed in clinical research as well since they can help in determining tumor types, in viral pathogenesis and also in studying neural development. miRNA-based therapies for human heart diseases are also in clinical trials. Graduate student Roma Broadberry uses cryo-electron microscopy (cryo-EM) to explore and investigate how viruses replicate and spread. Protein structure is crucial to understanding how a virus replicates and can help identify new targets for drugs, therapeutics and even shed light on neurodegenerative diseases. “If we can understand the structure as well as we can, we can improve our medicinal approaches,” Broadberry says. “Cryo has become this incredibly popular, accelerated approach to solve protein structures.” As a member of the Tim Grant Lab, her focus is on developing cryo-EM methodologies to determine the structure of biomolecules, improve sample preparation, computation and processing needs.
