Ludwig Lausanne's Melita Irving, recent PhD graduate Evangelos Stefanidis and colleagues report in the Journal of Clinical Investigation a novel two-pronged immunotherapy strategy to simultaneously drive T cell and macrophage attack on tumors. Their approach modifies T cells equipped with affinity optimized T cell receptors to additionally secrete a factor that stimulates macrophage attack on cancer cells. This approach, combined with clinical antibodies like Avelumab and Cetuximab, significantly boosts the immune system's ability to eliminate cancer cells. In a study funded by the Human Tumor Atlas Network (HTAN) and the Cancer Systems Biology Consortium (CSBC), Dr. Thomas Walle , Dana Pe'er, et al. developed Spectra, an algorithm for the supervised discovery of interpretable gene programs from single cell data. They showed that Spectra outperforms existing approaches in deciphering tumor immune contexts, including the identification of cellular mechanisms associated with responses to immunotherapy. Congratulations this talented and dedicated group of scientists Aileen Li, PhD, Jessica B., Jia Lu, Quinn Walker, Rowena Ellsworth, Hajime Hiraragi, Bijan Boldajipour, Purnima Sundar, Shobha Potluri, Gary Lee, Omar Ali, and Alexander Cheung on the publication in Scientific Reports that describes our novel Stim-R T cell activation technology. CAR T cells generated using our Stim-R technology showed improved potency, expansion and cytokine production compared to standard T-cell activation reagent control. In addition, characterization of Stim-R derived CAR T-cells revealed retention of a stem-like cell population and downregulation of exhaustion-associated gene sets.  This innovative technology is incorporated in LYL119, our next generation ROR1 CAR T cell product candidate entering a Phase 1 development in a clinical trial that will initially enroll patients with ROR1‑positive platinum‑resistant ovarian cancer or endometrial cancer. https://lnkd.in/gha5VchW

We are proud to advance rare disease research and patient outcomes. In collaboration with The National Institutes of Health (NIH), National Center for Advancing Translational Sciences (NCATS), and the GW Computational Biology Institute, CVP recently published its work analyzing national insurance claims data to better understand diagnosis, treatment, and costs associated with 14 rare diseases. Key findings: ▶️ Significant variability in prevalence and costs across US regions and between urban/rural areas. ▶️ Pharmacy costs increased with new treatments, while non-pharmacy costs (inpatient, outpatient, and/or physician costs) decreased. ▶️ Challenges in prevalence estimation are due to limited data and coding issues. Today we presented a poster highlighting preclinical data demonstrating anti-tumor activity of RGT-61159, our potent, selective oral small molecule inhibitor of the MYB oncogene, to treat acute myeloid leukemia (AML) at the 66th American Society of Hematology (ASH) Annual Meeting. These data confirm the role of MYB as an oncogenic driver across a broad AML patient population and support the company’s plans to advance RGT-61159 into a Phase 1a/b clinical trial to further explore its potential as a treatment for AML. #Ang-2 correlated positively with BNP, cTnI, sCr, E/e′, and the extracellular water (ECW)/intracellular water (ICW) ratio (ECW/ICW). After 24 months, a deterioration in renal function was observed in 47 patients (58%). In multivariate regression analysis, both VCAM-1 and Ang-2 showed independent influences on risk of renal replacement therapy initiation. In a Kaplan-Meier analysis, 72% of patients with Ang-2 concentrations below the median (3.15 ng/mL) survived without dialysis for two years. Such an impact was not observed for GFR, VCAM, CCP, VEGF-C, or BTP. Endothelial activation, quantified by plasma levels of Ang-2, may play a key role in GFR decline and the need for dialysis initiation in patients with CKD 3b, 4, and 5. https://lnkd.in/dR-DBsRC

We recently shared our work on using JESS automated western blotting to track phospho-proteins in a signaling pathway. By leveraging biomarkers, we developed a screening cascade that identified drug compounds with high specificity and potency, crucial for advancing therapies with minimal side effects. This approach ensured the targeted inhibition of a signaling pathway branch, offering a more refined and effective therapeutic option. Our findings pave the way for successful transitions from in vitro to in vivo studies, reinforcing the drug development pipeline.
