A large team of microbiologists, pathologists and infectious diseases specialists affiliated with several institutions in the U.S., including University of Pennsylvania Perelman School of Medicine, University of Pennsylvania, Children's Hospital of Philadelphia, The University of Texas Medical Branch, Polytechnique Montréal, Acuitas Therapeutics, Inc., has developed an #mRNA #vaccine that has thus far been found able to protect mice against intestinal #Clostridioidesdifficile bacterial #infection. https://lnkd.in/eJyuxcyx In their paper published in the journal Science, the group describes how they used mRNA technology to design a multipronged vaccine to target several proteins that are needed by the bacteria to cause gut ailments and how well it has done in testing with mouse models. Prior research has found that patients in hospitals who are given antibiotics to treat various infections are left with vulnerable bowels after the good bacteria in their gut is destroyed. This leaves them open to bacterial infections such as #Cdifficile, which infects the gut and releases toxins that cause a variety of ailments including #diarrhea, severe cramping and in some cases death due to #sepsis. https://lnkd.in/eJyuxcyx Medical researchers have been looking for ways to combat these bacteria, but thus far, the microbes have been able to evolve faster than the drugs used to kill them. In this new effort, the researchers used lessons learned from research involved in developing vaccines for the virus that causes COVID-19 to develop one that could effectively fight C. difficile—at least in mouse models. The messenger RNA (mRNA) in vaccines used to fight #COVID19 involves getting human #Tcells to create proteins that indirectly interrupt the virus's ability to attach to the cells they are trying to infect. The research team used mRNA to get multiple components of the immune system (immunoglobulins, T cells and antibodies) to produce several types of proteins that could be used to fight C. difficile during different parts of its life cycle, ensuring that it could be effective in eliminating the bacteria from a host. In testing their vaccine, the researchers found it to be very effective—all the mouse models given normally lethal doses of C. difficile recovered, while those in a control group all died. Finishing off our #MeetThePostDoc series hearing from postdoc, Dr. Nisreen Al-Namnam. What led you to pursue a postdoc, and how has your journey shaped your research and career path? I pursued a postdoc driven by my passion for research and teaching in medical sciences. Fascinated by bone regeneration and healing during my PhD, I sought to explore the field further. The postdoc experience significantly impacted my career by enhancing my technical, leadership, and project management skills. It also expanded my research network, allowing me to collaborate with experts and secure funding. More importantly, it helped clarify my academic goals and refine my research vision through interdisciplinary collaborations, ultimately shaping my career path. Can you share a memorable moment or achievement during your postdoc experience that made you proud of your work? One of the most memorable moments during my postdoc was winning a prestigious fellowship to represent BORS (British Orthopaedic Research Society) while traveling across North America. This opportunity allowed me to present our research on the effects of surgical instruments on bone tissue to top academic institutions and conferences. Engaging with renowned experts, receiving valuable feedback, and learning about innovative developments in the field was both inspiring and rewarding. It was a proud achievement that expanded my professional network and reinforced the significance of our work in advancing orthopaedic research and improving surgical outcomes. What’s one piece of advice you would give to early-career researchers or anyone considering a postdoc? One key piece of advice I would give to early-career researchers or those considering a postdoc is to think ahead from day one and work strategically toward your long-term goal, whether that’s securing a permanent position, obtaining grants, or earning a fellowship. Stay focused on your passion and actively pursue opportunities that align with it. Don’t miss any chance to learn new skills—be it through collaboration, mentorship, or hands-on experience, and be proactive in building your expertise and network. In a study funded by the Human Tumor Atlas Network (HTAN) and the Cancer Systems Biology Consortium (CSBC), Dr. Thomas Walle , Dana Pe'er, et al. developed Spectra, an algorithm for the supervised discovery of interpretable gene programs from single cell data. They showed that Spectra outperforms existing approaches in deciphering tumor immune contexts, including the identification of cellular mechanisms associated with responses to immunotherapy. 🔬 A new review by researchers from Université Claude Bernard Lyon 1 describes next-generation prospects for antibody–drug conjugates (ADCs). In the coming months, three ADCs should receive marketing authorization to treat various forms of NSCLC in adults: datopotamab deruxtecan, patritumab deruxtecan, and telisotuzumab vedotin. • Thirteen antibody–drug conjugates (ADCs) are currently commercially available. • The drug:antibody ratio must reflect the precision of the conjugation process used. • In addition to mastering linker design and payload selection, target diversification remains crucial. We are thrilled to share the presentation of Stephanie Chrysanthou, PhD, our R&D Manager at Integrated Genomics Operation Memorial Sloan Kettering Cancer Center, on detection of germline alterations in solid #cancer patient samples by adaptive sampling 🙌 presented at the Nanopore Community Meeting conference in Boston! Our core has closely partnered with the Molecular Pathology & Diagnostics lab to benchmark Adaptive Sampling long read #sequencing for detecting germline #mutations against prior MSK-IMPACT profiles generated by hybridization capture short-read sequencing. Adaptive Sampling is a promising approach due to reduced technical complexity and TAT, there is no need for sample batching and it facilitates the resolution of large complex structural #variants. Due to the live #basecalling capacity of the #nanopore technology, the sequencing can be stopped if a fragment does not match the uploaded target .bed file, thus enriching on-target fragment #sequencing. Briefly, our on-instrument data workflow includes: 1) filtering of the raw .pod5 files based on the on-target decision, 2) SUP basecalling, 3) GRCh38 alignment, 4) SNV/SV calling using #EPI2ME and #SEVERUS pipelines.  On a routine basis, this workflow, from #hmwDNA extraction to data analysis, takes 3.5 days. Congratulations to Matthew R. Sullivan, et al. & Tania Konry from Northeastern University on their paper publication “Characterizing influence of rCHOP treatment on diffuse large B-cell lymphoma microenvironment through in vitro microfluidic spheroid model”! The study introduces a hydrogel spheroid-based microfluidic model for screening lymphoma treatments, aiming to address the challenges in improving patient survival rates for diffuse large B-cell lymphoma (DLBCL). Incorporating VitroGel® RGD into the hydrogel composition enhances cellular viability within the 3D culture environment. The RGD-rich hydrogel promotes cell adhesion, leading to increased viability of DLBCL and natural killer (NK) cells compared to alginate alone. Additionally, the rheological properties of the hydrogel more closely resemble those of metastatic lymph nodes, enhancing the model's physiological relevance.
