I can see a real network effect happening at Aanika Biosciences, Inc. There are oporuntitoes forming for cross-selling, partnerships and co-development between our customers. And in the middle, sits Aanika de-risking it all. Aanika has become a clearing house for complex technologies and products first for agriculture but I can see this growing much beyond that. #networkeffect

Wuxi's exit from the US CDMO space will leave a gap. Currently, Indian CDMOs look like the most likely candidates to fill it. A key feature of Indian CDMOs' strategy has been to remain independent from Chinese suppliers, mitigating the risk of disruption. As other CDMOs scramble to find new suppliers, India will emerge as a key replacement for companies like Wuxi. However, there will still be opportunities for European organisations if they act fast. Bristol Myers Squibb pays $110M to form T-cell therapy pact, helping Prime Medicine, Inc. buy time to advance prioritized pipeline Bristol Myers Squibb is paying Prime Medicine $110 million upfront to develop reagents for ex vivo T-cell therapies. Prime, which could receive a whopping $3.5 billion in milestones, disclosed the deal alongside details of a pipeline prioritization intended to extend its cash runway into the first half of 2026. BMS is partnering with Prime to further its work to unlock the potential of cell therapy in immunology and oncology. In return for the upfront payment, which is split evenly between a cash payment and equity investment, Prime will design optimized Prime Editor reagents for a select number of targets. What’s new from PacBio ? The new SPRQ chemistry reduces sequencing cost - now you get a $500 human genome! I’ll talk about how this affects Kinnex in a separate post but here’s on how it affects WGS. First, it’s the same HiFi data - no change in accuracy or read length - but with reduced DNA input to 500ng and 30% more yield. This means for human WGS you can now get two 20x genomes on one Revio SMRT Cell for $500 each. But why 20x? We’ve shown that at 20x HiFi you get better SNV and indel calls than Illumina or ONT. The new PacBio Revio SPRQ chemistry increases yield by 33% - what does this mean for Kinnex RNA sequencing? And what data is there to show it? First, SPRQ does not change accuracy or read length. You can expect the same captured transcript lengths. What changed is yield - for Kinnex full-length RNA kit, the expected yield is now 60M reads per Revio SMRT Cell - a 50% increase from the previous 40M standard. This means to get ~10M reads per sample (our std recommendation for bulk transcriptome), instead of doing 4-plex per Kinnex library, you can now do 6-plex! What does this mean for cost per sample? It means: At 10M read depth (6-plex), it is now $325 USD per sample (at list price) from cDNA generation to sequencing. At 5M read depth (12-plex), it is now $243 USD per sample. We've released a matching UHRR(2024) Kinnex FL library run w Revio original vs Revio SPRQ. Both runs hit beyond spec. There's no bias in using Revio original vs SPRQ chemistry. It's the same Kinnex library, after all, and just more data. The isoform abundances correlate very well. The two UHRR2024 datasets have been added to the Kinnex FL URL. Automation of well plate and culture tube processes are now well established for microbial and all types of mammalian + insect cell culture systems. Automation-driven capacity gains and culture-cost reductions once thought limited to mainstream biologics (transient + stable CHO fed-batch) are now possible with technically challenging AAV gene therapy, CHO perfusion, CAR-T applications and large volume cultures (300ml-1,800ml)… if you know what you’re doing. Not all scaling factors are created equal: matched kLa, consistent energy dissipation rates (shear) and controlling evaporative losses are important (among other factors) – but each application may have different priorities. Tremfya, Carvykti sales grow as J&J prepares for Stelara patent loss Biosimilar competition is expected in early January for the company’s second highest-selling drug, putting greater focus on newer products’ market trajectory. Dive Brief: Sales of Johnson & Johnson’s cancer therapy Carvykti and immune disease drug Tremfya grew strongly during the third quarter, the pharmaceutical company reported Tuesday, as new approvals helped broaden the two medicines’ market reach. The growth from Carvykti and Tremfya comes as J&J prepares for biosimilar competition to its second best-selling drug, Stelara, in January 2025. Biosimilars have already arrived in Europe and, alongside larger-than-expected insurer rebates, drove Stelara’s third quarter sales down 7% compared to last year. While analysts viewed the performance of J&J’s medical device division as underwhelming, J&J was still confident enough in its outlook to raise operational sales guidance for the full year slightly.
