Roche's recent decision to offload their Vacaville facility highlights the increasing importance of cell line productivity in biomanufacturing strategy. According to Dan Stanton of Bioprocess Insider, Roche is moving towards more high titer processes and products with lower volume demands. (You can read the article here https://lnkd.in/erRx597k) Agile manufacturing strategies are enabled by cell line productivity, which allows for the same amount of drug substance to be produced in smaller bioreactors. This, in turn, allows for smaller clean rooms and lower running costs. Just - Evotec Biologics' J.CHO High Expression System cell lines, grown in perfusion bioreactors, produce over 4 g/L of antibody each day, equivalent to around 30 g/L in fed-batch mode (https://lnkd.in/eexeJrQA). This level of productivity allows for commercial quantities of antibody to be manufactured in 1000L single-use bioreactors in our modular J.POD facilities. As a result, large-scale stainless-steel facilities are becoming redundant. UC Irvine study reveals circadian clock can be leveraged to enhance cancer immunotherapy Optimizing individual time-of-day delivery offers new avenues for prevention and treatment Irvine, Calif., May 28, 2024 — A multidisciplinary research team at the University of California, Irvine has revealed that the circadian clock – the biological pacemaker that governs daily rhythms in physiological processes, including immune functions – can be leveraged to enhance the efficacy of checkpoint inhibitor cancer therapy. Checkpoint inhibitors block different proteins from binding to tumor cells, allowing the immune system’s T cells to kill the tumor. The study, published online today in the journal Nature Immunology, provides deeper insights into the intricate relationship among the circadian clock, immune regulation and tumor development and found that a therapeutic approach optimizing time-of-day delivery based on an individual’s unique circadian patterns offers new avenues for prevention and treatment. “Disruption of the internal biological pacemaker is an inherent aspect of modern society that may contribute to the rising incidence of many cancer types. We found that proper regulation of circadian rhythms is necessary to suppress inflammation and support peak immune function,” said corresponding author Selma Masri, UC Irvine associate professor of biological chemistry. “Understanding precisely how circadian disruption promotes disease progression could lead to behavior modification to reduce cancer risk.” Team members used an advanced single-cell RNA sequencing technique in a genetic model of colorectal cancer and identified clock-dependent changes controlling the number of myeloid-derived cells that suppress T cell activation. They discovered that disruption of the internal clock in the epithelial cells lining the intestine alters secretion of cytokine proteins, leading to heightened inflammation, increased numbers of immunosuppressive myeloid cells and cancer progression. I am excited to announce that one of Access Discovery Bio’s first clients is Cache DNA based out of San Carlos, CA. Cache has an exciting new technology that allows for the “Caching” of DNA and RNA, allowing them to be stored at ambient room temperature. Caching can be done on a small scale with various tubes or can be completely integrated into existing automated technologies that handle plates for working at scale. This is a profound development in my eyes after decades of both working in biorepositories and shipping frozen samples all over the world. I have personally witnessed sample evaporation and loss despite long term cold storage. I’ve seen the dramatic rise in costs not only to create and maintain freezer farms but also to safely ship frozen samples which can often dip into the thousands for a single shipment. I have personally been on call and had to travel into a lab at 2am to recover samples in a failed freezer or during a power outage. I have also worked in the lab having to use various techniques to properly freeze or thaw samples to work with them. For many reasons, I see this as a transformational technology. Who doesn’t like to save massive amounts of money? Everyone does of course and this comes with the added benefits of reducing carbon footprints and helping the environment. Michael Becich and his team at Cache are a great group of scientists who have created something truly beneficial to basic science. Cache is presently offering free demo kits to select customers while supplies last. Element Biosciences has raised an additional $277 million in private financing to support commercialization of its benchtop DNA sequencer and an instrument launch as the company takes on market leader Illumina. Founded in 2017, Element is part of the race to drive down the cost of DNA sequencing. It’s also pushing into multiomics, which includes RNA and protein data. The haul brings Element’s fundraising total to $680 million. I was looking at some of the up and coming biotech hubs in the U.S. According the an article in Drug Discovery & Development, Phoenix, Arizona is a rising star. "The bustling city is home to Phoenix Medical Quarter, a 100-acre hub set in the city’s midtown district, which aims to compete with top global medical research centers. The Mayo Clinic is also building its Discovery Oasis, a 120-acre medical and biotech research hub is strategically located between Mayo Clinic’s hospital campus and Loop 101 in north Phoenix." Super proud of the team and product we’ve built. When I first learned about Weave and the potential to accelerate time to trial for drugs, I was intrigued. I saw the same issues in other domains. Both in rocketry and carbon markets. One party is trying to bring innovative solutions to market while demonstrating the safety and accuracy of what they’re making. The burden of proof is on the makers. Third parties have to sift through documents and data to verify the integrity of what’s been done and green light moving forward. What’s clear is that organizations need better ways to organize and collaborate around these documents, prepare them efficiently, and deliver them easily. Regulators need better ways to sift through the hundreds of pages of documents to easily answer “is this safe?”. Its not surprising that documentation preparation and review can become a significant bottleneck in drug development. LLMs offer an unprecedented opportunity to change how we collaborate around data in safety critical applications. Investigational New Drug (IND) applications are extremely complex. We are providing a way to simplify them that and interact with them to accelerate the whole process. What excites me most is that we get to build an AI-first application based off the latest advancements that really transform the user experience. We get to reimagine a lot of functionality on how to leverage AI first then put the people in the driver seat to accelerate what they can do individually. Then we get to think of ways how we can support organizations, whether it’s automated reviews, suggested edits, auto updating generated content, or something we haven’t thought of yet. We get to help people and organizations accelerate their regulatory obligations without compromising the integrity of the results. They get through the process faster.
