By combining the single-cell analysis approach of #flowcytometry with the precision of #massspectrometry, #masscytometry enables high-parameter analysis while generating little to no signal overlap. This eliminates the need to correct fluorescence spillover and further enables the use of statistical techniques to facilitate screening drug candidates and assessing on- or off-target effects post-treatment. #Livercancer is the third leading cause of cancer-related mortality worldwide. To improve immunotherapy response rates, the ability to accurately identify responders from non-responders is crucial. Scientists from Huazhong University of Science and Technology used single-cell CyTOF™ technology and mIHC analyses to generate a comprehensive and dynamic immunological atlas associated with response and prognosis in advanced liver cancer patients who underwent treatment with immune checkpoint inhibitors. #CyTOF assisted in identifying predictive and prognostic #biomarkers based on three baseline characterizations in a cohort of 24 patients, supporting patient stratification and fostering new response monitoring strategies. Mass cytometry is advantageous because of its ability to precisely and accurately distinguish between reporter molecules. By using atomic mass measurements, mass cytometry avoids autofluorescence signals from cells and tissues, enables fast panel design since all antibodies are used at once and facilitates barcoding to multiplex samples in large-scale acquisitions.
