2024 was a pivotal year in the evolution of Parabilis Medicines and our mission to deliver extraordinary medicines for patients. We are now called Parabilis Medicines, and were formerly FogPharma. This new name embodies our vision of shooting for the moon with our aspiration to create transformational medicines, yet grounded in a strong sense of pragmatism and execution. 2024 has been an amazing year for us – progress along every dimension of the company. The top highlights demonstrating our substantial progress include: • We welcomed many extraordinary colleagues to our team, and we were recognized by the BostInno Fire Awards, Biotech Week Boston Awards, and Scrip Awards for our exciting progress and impact within the biotech ecosystem. • We advanced FOG-001, our first-in-class β-catenin:TCF inhibitor, in its clinical development. FOG-001 is the first of its kind – a designed alpha-helical peptide that our preclinical studies have shown enters tumor cells and inhibits a protein-protein interface that has been considered undruggable for decades due to lack of any small molecule binding cavity. • We significantly advanced our discovery pipeline with multiple programs now moving towards Development Candidate nomination. Each of our programs is unique in the industry, and each targets an intractable protein that is a critical driver of cancer. • We established an important co-create partnership with ARTBIO to discover and develop HEARTs (Helicon Enabled Alpha Radiotherapies) that are intended to deliver cancer-killing alpha particles via a Helicon-chelated Lead-212. • We invested deeply in and are continuously evolving our Helicon platform that created FOG-001. This platform now integrates cutting-edge AI and several multiplexed, advanced, and automated experimental technologies. • Finally, we successfully closed a $145 million Series E financing backed by an industry-leading syndicate of investors. With nearly 30 years of biopharma R&D experience, Anandan has deep expertise managing high-performing teams, fostering cross-functional collaborations, and building strategic partnerships. Anandan brings specialized expertise in small molecules and peptides. One special feature that sets Helicons apart from other peptide therapies is their tunability. Helicons have tunable clearance characteristics to achieve short or long plasma and tissue residence times, and tunable exposure to different tissue types. This makes Helicons well-suited for the creation of #radiopharmaceuticals for treating #cancer. By co-developing Helicon-enabled alpha radioligand therapies (HEARTs) with ARTBIO, we’re harnessing Helicons’ tunable characteristics to enable the targeted delivery of lead-212. We were thrilled to recently welcome Mohammed AlQuraishi from Columbia University to Parabilis for a discussion about the state of the art in computational protein design today. Dr. AlQuraishi is a world expert in systems biology who recently played a central role in the development of OpenFold, an open-source AI model for predicting the 3D structures of proteins from their sequences. Following Dr. AlQuraishi’s seminar we had an engaging roundtable discussion about the leading edge of #bioAI modeling, such as prediction of complexes and mutant variants, as well as the current and future role of these models in Parabilis’s target identification and #drugdiscovery. It was such a delight to be invited to participate in this year’s Molecular Machine Learning Conference (#MoML2024) organized by MIT Jameel Clinic. The event brought together a group of innovators at the forefront of applying #AI, machine learning and physics-based modeling to discover new drugs and solve challenging problems in health and medicine. CEO Mathai Mammen, M.D., Ph.D. presented about how we at Parabilis are breaking new ground in applying AI and data science to discover and develop Helicon therapeutics.
