IMRX’s VP of Clinical Pharmacology, Jan de Jong, presented interim Phase 1 pharmacokinetic & pharmacodynamic data on IMM-1-104, a novel oral Deep Cyclic Inhibitor of the MAPK pathway, at the 2024 ACoP Conference. These PK/PD modeling data, combined with Phase 1 safety and activity data, suggest that 240 mg and 320 mg are viable doses to promote Deep Cyclic Inhibition of the MAPK pathway. We have established a reporter mRNA-based screening platform for in vivo evaluation of the functional payload delivery of our customized LNPs. Our expert, Bart Vanderborght, is happy to share his knowledge with you: The fast-expanding field of nucleic acid-based therapeutics has the potential to unlock access to a broad range of specific disease indications. To achieve this however, it is vital that the nucleic acids are delivered to the target organ, and, in that organ, to the target cell type. At etherna, we have established a reporter mRNA-based screening platform for in vivo evaluation of the functional payload delivery of our customized LNPs. This platform enables bioluminescence imaging-mediated assessment of the 𝐛𝐢𝐨𝐝𝐢𝐬𝐭𝐫𝐢𝐛𝐮𝐭𝐢𝐨𝐧 𝐭𝐨𝐰𝐚𝐫𝐝𝐬 𝐚𝐧𝐲 𝐨𝐫𝐠𝐚𝐧 𝐨𝐟 𝐢𝐧𝐭𝐞𝐫𝐞𝐬𝐭 following administration of reporter mRNA-formulated LNP to mice (upper part of the figure). In addition, through the integration of the assessment of reporter protein expression in our flow cytometric analysis, we can determine the 𝐜𝐞𝐥𝐥𝐮𝐥𝐚𝐫 𝐭𝐫𝐨𝐩𝐢𝐬𝐦 𝐭𝐨 𝐬𝐩𝐞𝐜𝐢𝐟𝐢𝐜 𝐜𝐞𝐥𝐥 𝐭𝐲𝐩𝐞𝐬 𝐨𝐟 𝐢𝐧𝐭𝐞𝐫𝐞𝐬𝐭 in a broad range of tissues (lower part of the figure). The DRUP trial, a pivotal study in precision oncology conducted across the Netherlands, has demonstrated significant strides in using off-label therapies based on patients’ genomic profiles. With over 1,500 patients treated, the DRUP trial has shown a sustained clinical benefit rate of 33%, offering options for those with advanced cancers. Key successes include high response rates in cohorts like the nivolumab group for MSI tumors, which showed a clinical benefit rate of 63%. These findings not only underscore the potential of precision oncology but also pave the way for broader application and reimbursement of these therapies. As DRUP expands across Europe, its collaborative framework promises to enhance our understanding of cancer treatment and foster equitable access to precision medicine. This ongoing work not only contributes to the scientific community but also to the patients who stand to benefit the most from these advancements. We are launching a targeted attack on prostate cancer! My company, CimCure B.V., in collaboration with SwiftPharma (Ghent, Belgium) and Amsterdam UMC, has secured a €1.1 million EU Eurostars project to develop a multi-targeted vaccination-based immunotherapy for prostate cancer. The project, titled "A sustainable first-in-class multi-targeted immunotherapy for the treatment of prostate cancer," will utilize an innovative production method using tobacco plants (Nicotiana benthamiana, see image below). This sustainable “green” approach eliminates the need for bioreactors, culture media, electricity, maintenance instruments and chemicals, relying solely on soil and sunlight. Founder and MITIGATE trial principal investigator John Stone MD, MPH shares: "Today, fifteen years after we first used a B cell depletion strategy in IgG4-RD, we now have confirmation through MITIGATE – the first such international trial ever performed in IgG4-RD – that this treatment approach of B cell depletion in IgG4-RD is effective. Inebilizumab reduced the risk of disease flare by 87% compared to the standard treatment arm. Our orphan disease now has a therapy that is proven to work." Using bioengineered bacteria to produce and deliver therapeutics in the body has tremendous potential. However, a significant concern with using such organisms is their potential to colonize the host, making it impossible to ever shut off delivery of the therapeutic. FRI executive committee member Jan-Peter van Pijkeren and his lab created a strain of lactic acid bacteria that contains mutations in nine (known and putative) adhesins (bacterial molecules that allow bacteria to attach to a host). In collaboration with FRI executive committee member LAURA KNOLL and her lab, they showed that the “nonuple” mutant had reduced adhesion ability to enteroid epithelial cells compared to the wild-type strain.
