ALS Community, Scientists and Researchers - Look out! At Axol Bioscience we continue to build tools, infrastructure and assays that support “Better Human Disease Modeling” decrease time and risk of Drug Discovery in ALS and ultimately improve patients lives. Massive THANK-YOU to the Scottish Government for their continued support of Axol Bioscience through the awarding of this SMART Grant. The key objectives of this project are to: 1. Develop a quality manufacturing system for iPSC-derived ALS relevant cell types 2. Characterise and validate the cells using ALS relevant end points; TDP43 aggregation, Neurofilament Light elevation and Cryptic Exon Sequencing 3. Develop and characterise an ALS co-culture model which includes motor neurons, microglia and astrocytes 4. Commercialise an easy-to-use model system to drive forward ALS drug discovery The grant is part of a broader push to scale the impact of Scotland’s innovation strengths, with a focus on projects that contribute to economic transformation. It's great that iPSCs have been recognised by the Scottish Enterprise to be a part of this cause. Such an exciting clinical milestone for Tryptamine Therapeutics (Tryp Therapeutics) with the company releasing clinically significant data from its second Phase 2a clinical trial utilising TRP-8802 (oral-psilocybin), this time, in Fibromyalgia Syndrome. For the pharmaceutical industry, from "New Chemical Entity" (NCE) status to completion of Phase 2a studies can take anywhere between 6 - 10 years and cost between US$200M - US$1B. Since Tryp Therapeutics had spent the last 4 years building data and IP around TRP-8802/8803 as well as selecting novel indications of high unmet clinical need, the company entered the ASX with a significant head start on potential competitors and will hold three levels of IP protection - molecule, IV-infusion delivery and novel indications. Today marks a pivotal milestone in the history of #TCR therapeutic development with the first ever TCR #celltherapy approved for a solid tumor. Huge congratulations to Adaptimmune for the approval of #TECELRA which is now available for adults with advanced MAGE-A4+ synovial sarcoma. I am excited to share Sunflower’s newest article published in BioPharm International, which I had the privilege of authoring alongside my wonderful co-authors, Laura Crowell and Stacy Martin. In this piece, we explore the productivity of cultivation processes, comparing different host organisms and manufacturing processes. We found that #continuousfermentation processes using yeast deliver the highest achievable space-time yields for recombinant protein production, resulting in more protein for less space and time. Ultimately, considering space-time yield as a critical metric when assessing upstream process designs can lead to lower biomanufacturing operating costs. 🗞️Have a look at our latest case study in which we helped Medicines for Malaria Venture (MMV) generate an antimalarial with reduced toxicity in less than an hour. 🌩️MMVs lead candidate in their aryl piperazine series exhibited high CYP450 activity which may cause adverse interactions with other drugs in humans. 🌤️Using DeepMirror, we generated 1,000+ compounds and quickly identified a compound with high antimalarial activity but lower CYP450 activity than the lead. This compound was subsequently validated experimentally by MMV testing entities. 🤷🏻‍♀️Why does this matter? 🚀 New Blogpost Alert!🚀 Excited to announce our latest blogpost in which we benchmark different generative algorithms (e.g. #claude and #REINVENT) for their capabilities to generate novel and valid molecules. Turns out most modern generative algorithms are pretty good and can help chemists if paired with good models for activity and ADMET! Highlights: 🤖 Claude, a large language model, appears to be an ok molecule generator ✍️ Classic medicinal chemistry driven approaches are competitive against more modern AI driven approaches such as #REINVENT 👏 All algorithms are good at scaffold hopping, i.e. generating molecules that are very different from original ones! Pleasure to announce - Glafabra Therapeutics is now launched by Devinebio. Their lead asset is is a cell therapy using genetically-modified hematopoietic stem cells from the patient, the GLA-H program. Patients get a constant supply of missing enzyme, alpha-galactosidase A. High durability at 5 years and possibly even longer. Yet, also in development is GLA-T, that substitutes in modified T cells as an easier modality to administer to the patient. The Cyclical Dance of Risk in Biotech: Assets In, Platforms Out? The "assets-in, platforms-out" sentiment in biotech is rearing its head again. This trend, where investors favour late-stage assets over early-stage platforms, isn't new. It pops up during "risk-off" periods, like the post-genomics bubble burst (2002-2005) or the Great Recession (2009-2012). We see it happening again after the pandemic market correction. Is this shift good for innovation? While understandable during economic jitters, prioritizing late-stage assets can starve the early-stage platforms brimming with potentially groundbreaking ideas.
