🧠 Publication in #NatureCommunications from the #GLORIA #glioblastoma cooperation across six universities, lead by Frank A. Giordano from the #DKFZHector Institute in Mannheim. They investigate the effects of Olaptesed Pegol (#NOX-A12, TME Pharma) combined with #radiotherapy in #glioblastoma patients. 🔬 “Our approach focuses on the tumor microenvironment rather than just the radiation effects on tumor cells. NOX-A12 inhibits new blood vessel formation, a process triggered by residual tumor cells post-radiotherapy,” explains Prof. Dr. Frank Giordano. Glioblastomas regenerate by promoting new blood vessels via signaling molecules like CXCL12. NOX-A12 blocks CXCL12, disrupting this process. The combination with radiotherapy enhances its efficacy. 💡 Early Phase I/II study results in patients resistant to standard therapy show promising safety and efficacy data. Extensive tissue analysis by @University Hospital Bonn suggests a correlation between high CXCL12 levels and better NOX-A12 response, warranting further studies. Gene therapy has evolved as a potential therapeutic option for treating Alzheimer’s disease, owing to its rapid advancement over the recent decade. Small interfering ribonucleic acid has recently garnered considerable attention in gene therapy owing to its ability to down-regulate genes with high sequence specificity and an almost limitless number of therapeutic targets, including those that were once considered undruggable. However, lackluster cellular uptake and the destabilization of small interfering ribonucleic acid in its biological environment restrict its therapeutic application, necessitating the development of a vector that can safeguard the genetic material from early destruction within the bloodstream while effectively delivering therapeutic genes across the blood-brain barrier. Nanotechnology has emerged as a possible solution, and several delivery systems utilizing nanoparticles have been shown to bypass key challenges regarding small interfering ribonucleic acid delivery. By reducing the enzymatic breakdown of genetic components, nanomaterials as gene carriers have considerably enhanced the efficiency of gene therapy. Liposomes, polymeric nanoparticles, magnetic nanoparticles, dendrimers, and micelles are examples of nanocarriers that have been designed, and each has its own set of features. Furthermore, recent advances in the specific delivery of neurotrophic compounds via gene therapy have provided promising results in relation to augmenting cognitive abilities. In this paper, we highlight the use of different nanocarriers in targeted gene delivery and small interfering ribonucleic acid-mediated gene silencing as a potential platform for treating Alzheimer’s disease.

Read our new open access publication by Palle Helmke, Barbara Füzi (former group member) and Gerhard Ecker on improving the drug development process! Development of a new drug currently takes 10–12 years with costs of around 2 billion EUR. The two main reasons for failures comprise lack of efficacy and unforeseen toxicity. For the latter, a standard process pursued to minimize the risk is the so-called toxicological read across. Briefly, toxicologists query the available literature and databases for compounds, which are structurally similar to their development candidate in order to retrieve information on potential hazards. In addition, computational models might be applied which are either trained for a single protein, such as hERG or P-glycoprotein, or for a respective in vivo endpoint (cholestasis, steatosis, drug-induced liver injury (DILI), …). In both cases, a proper ‘description’ of the compound of interest is key for the predictive ability of the models. Congratulations to Tubulis GmbH! The company announced that the U.S. Food and Drug Administration has granted Fast Track designation to its lead antibody-drug conjugate (ADC) TUB-040 for the treatment of patients with platinum-resistant ovarian cancer. TUB-040 is a NaPi2b-targeting Exatecan ADC based on Tubulis’ proprietary P5 technology with superior biophysical properties that demonstrated effective and durable responses in a range of preclinical models, including ovarian cancer. Source: (Journal of separation science) Study compares lipidomes of 12 Panax herbal medicines using UHPSFC/IM-QTOF-MS. Method resolves ginseng lipidome in 30 min, identifying 82 differential lipids. Root ginseng differs from stem/leaf/flower; red ginseng distinct from other roots. Findings aid in ginseng identification based on lipid profiles.
