Imaging-Based Screening Platform Assists Protein Engineering.
Cell Chemical Biology
We built a high-throughput screening platform that integrates high-quality fluorescence-based image analysis and robotic picking of bacterial colonies. It is highly adaptable and offers powerful, flexible, and low-cost instrumentation to accelerate many protein engineering projects. We demonstrate the power of the screening platform by optimizing a dim far-red-emitting fluorescent protein whose brightness increased several fold using iterative cycles of mutagenesis and platform-based screening.

The paper describes the High-Throughput Screening of red fluorescent proteins to generate a new far-red variant. It applies hypersomatic mutation to selectively mutate genes of interest at a high rate of mutation to generate a very large library of variants, for iterative FACS sorting to isolate beneficial mutations.
