🌟 New Drug-Conjugate Deals of June 2024 🌟 In June we witnessed 10 drug-conjugate deals. Below is a round-up of a few: - Biolojic Design, Ltd. & Merck KGaA: Entered a multi-target drug discovery collaboration. Biolojic Design received an upfront payment in the low double digits and could earn up to $375.3m in milestone payments. - ArriVent Biopharma & Alphamab Oncology: Teamed up to discover, develop, and commercialize #novel ADCs for cancer treatment. Alphamab could receive up to $615.5 million in combined upfront and milestone payments. - FREZENT Biological Solutions: Raised pre-seed #funding and disclosed their ADC and bispecific programs in early June 2024. - Day One Biopharmaceuticals & MabCare Therapeutics: Entered an exclusive licensing agreement for MTX-13. MabCare will receive £55 million upfront and could earn an additional $1.152 billion in milestone payments. MTX-13 is a preclinical ADC using #Camptothecin as a #payload. - Radiopharm Theranostics & Lantheus: Signed a licensing agreement for a #TROP2 targeting nanobody and a LRRC15 targeting Mab, with Lantheus paying an A$3m upfront fee. Both molecules are preclinical #radioimmunoconjugates. In June, we witnessed the introduction of 33 new drug-conjugate drugs, the initiation of 37 new trials, and the emergence of 10 new companies in the field. Additionally, there were 227 trial updates. A recently published study by Merck KGaA, Darmstadt, Germany, EMD Serono, Inc., Christian-Albrechts-Universität zu Kiel, and Technische Universität Darmstadt investigates bispecific NK cell-engaging antibodies (NKCEs) for cancer immunotherapy, focusing on the impact of antibody architecture on their efficacy. ✅ It highlights the importance of design, valency, and NKp30 epitope recognition in influencing NKCE characteristics. ✅ The research emphasizes the benefits of Fc domains for prolonged half-life and demonstrates that the tested NKCEs exhibit IgG-like pharmacokinetic profiles, indicating their clinical potential in cancer treatment. ✅ For simultaneous binding experiments as well as kinetic measurements the Octet®️ BLI technology was used. 𝗡𝗼𝘃𝗲𝗹 #𝗰𝗮𝗻𝗰𝗲𝗿 #𝘁𝗵𝗲𝗿𝗮𝗽𝗲𝘂𝘁𝗶𝗰 𝗲𝗻𝘁𝗲𝗿𝘀 𝗰𝗹𝗶𝗻𝗶𝗰𝗮𝗹 𝗽𝗵𝗮𝘀𝗲: Groundbreaking innovations by researchers at the Leibniz-Forschungsinstitut für Molekulare Pharmakologie (FMP) provided the basis for the development of an antibody-drug conjugate (#ADC) by the company Tubulis GmbH, which is now being used in a Phase I/IIa trial in tumour patients. Thanks to an innovative linker chemistry known as P5 technology, the active substance is delivered safely to its target and can also develop its effect over a long period of time. The beginning of a clinical trial just a few years after the discovery of the P5 technology is a major achievement that demonstrates the potential for applied research in an academic research institute. To combat cancer, there is a need for active substances that specifically target cancerous cells and destroy them without damaging the surrounding healthy tissue or other bodily functions, as is often the case with conventional chemotherapies. So-called antibody-drug conjugates (ADCs) can specifically attack tumour cells by delivering toxic substances directly to the tumour, bypassing healthy cells. ADCs have been used for many years but tend to have only limited efficacy. This is mainly due to the fact that active substances may fall off beforehand and fail to reach the targeted tumour cell. A team led by Prof. Hackenberger at the FMP has addressed this problem and developed the novel P5 technology, one of the central components of the ADC developed by Tubulis. For nearly 20 years, Hackenberger has been researching chemical processes that can be used to specifically functionalise proteins and antibodies. With his team at the FMP, he discovered reactions that provide stable conjugates of active substances and antibodies with excellent solubility properties. New publication from Danube Private University’s Life Science Technology Laboratory (LiST). Dr. Jakub Dostalek, Prof. Dr. Christoph Kleber and Dr. Roger Hasler have contributed to the research paper “Surface plasmon resonance biosensor with anti-crossing modulation readout” in the Journal Sensors and Actuators: B. Chemical. The paper presents a novel approach to surface plasmon resonance (SPR) biosensors, simplifying the label-free monitoring of biomolecular interactions. By using anti-crossing surface plasmon modes along a thin metal film atop a tailored multi-periodic grating structure, the sensor eliminates the need for optical matching with a prism and avoids probing through the liquid sample. This method enhances sensitivity and versatility in detecting SPR changes due to biomolecular binding-induced refractive index variations. The sensor, compatible with standard SPR readers and using mass-producible UV-nanoimprint lithography, demonstrates its potential by effectively characterizing antibody interactions with the cancer biomarker CSPG4. Last week we took some time to visit Professor Tobias Madl, Head of the Division of Medicinal Chemistry at the Medizinische Universität Graz. 😊 It was time to document the official handover of our BRAVE B-Curious nanoparticle analyzer. 🎉 👩‍🔬📝 👨‍🔬 This BRAVE B-Curious will make a contribution to research into metabolomics, e.g. how the molecular mechanisms of disordered proteins regulate signal transduction, intracellular transport and phase separation. These investigations help understand how modifications in signal transduction pathways affect the ageing process and the development of cancers and neurodegenerative illnesses. The University Medical Centre Mannheim at Heidelberg University in Germany will lead a Phase 1b/2 clinical trial for our lead drug SNT-5505, targeting high-risk myelodysplastic syndrome (MDS) and chronic myelomonocytic leukemia (CMML). Funded by a A$2.5 million grant from Deutsche Krebshilfe, this trial will commence in Q1 2025 and will include a dose-escalation phase followed by an expansion phase, using SNT-5505 in combination with 5-azacytidine. Running parallel to our Australian study for low/intermediate-risk MDS, this trial presents a combined market opportunity of approximately US$6 billion for SNT-5505. CEO Gary Phillips, said: "We are eager to further explore the potential of SNT5505 in addressing significant unmet needs in the treatment of MDS. The interest amongst academic clinical researchers is a direct response and validation of our long-term collaboration with Heidelberg University. By expanding our clinical trials to include both high-risk and low/intermediate risk MDS patients, we are positioning SNT-5505 as a potential treatment option for all MDS patients. This indication adds both clinical and commercial value to SNT-5505 where our ongoing myelofibrosis study is already fully recruited and will report interim data later this year." For everyone working in cancer research, I stumbled upon an article in the 'Deutsches Ärzteblatt' that I wanted to share. A new interactive tool called "Onkologie Monitor Rheinland-Pfalz" has recently been introduced, enabling a thorough examination of cancer registry information in Rhineland-Palatinate. Developed by the Institute for Digital Health Data (IDG), this online platform features up-to-date, verified data from the local cancer registry. Philipp Kachel, Managing Director of IDG, emphasized the importance of analyzing and utilizing oncology data to benefit patients while adhering to strict data protection guidelines. The dashboard consolidates more than 2.4 million records on diagnoses, treatments, tumor conferences, and patient conditions from residents or individuals receiving treatment in Rhineland-Palatinate. Users can filter and merge data from over 50 categories into as many as 15 age groups. It offers epidemiological indicators such as incidence rates, mortality rates, and prevalence rates, along with clinical specifics regarding disease stage, site of occurrence, and histology. Furthermore, users can compare Rhineland-Palatinate's data with national statistics from Germany. #𝗞𝗲𝘆 𝘁𝗼 𝗜𝗺𝗽𝗿𝗼𝘃𝗶𝗻𝗴 #𝗖𝗮𝗻𝗰𝗲𝗿 𝗧𝗿𝗲𝗮𝘁𝗺𝗲𝗻𝘁𝘀 𝗗𝗶𝘀𝗰𝗼𝘃𝗲𝗿𝗲𝗱: Some cancer drugs cause severe side effects because they are not working accurately enough. A team at the University of Würzburg led by biochemist Caroline Kisker has now discovered why. The small protein ubiquitin is involved in almost every cellular process in our body: it orchestrates the stability and function of the vast majority of proteins. When it binds to other proteins, they are often released for degradation. However, this labelling can also be reversed by special enzymes. The enzyme USP28, for example, is known to stabilise proteins that are important for cell growth and division – these can also play an important role in cancer growth.
