Heart and lung diseases are the leading causes of death worldwide with unmet treatment challenges that can only be tackled by deciphering the molecular biology underlying individual cellular phenotypes and intercellular as well as inter-organ cooperativity. Similarly, understanding of failed regulation in disease and its exploitation for novel therapies is lacking. These challenges in molecular medicine are addressed by the Cardiopulmonary Institute (CPI) a consortium of basic scientists, translationally-oriented researchers and clinicians at the universities of Frankfurt (GU) and Giessen (JLU) and the Max Planck Institute for Heart and Lung Research (MPI-HLR). This consortium is funded by the Excellence Strategy program of the German Research Foundation and is now seeking additional expertise in emerging fields

👏 Congratulations to Professor Mauro Giacca on receiving an honorary doctorate at the Goethe-Universität Frankfurt “Dies Academicus” ceremony, in recognition of his exceptional contributions to #Molecular #Biology. His groundbreaking work has significantly advanced the fields of gene therapy, stem cell research, cardiovascular disorders, and infectious diseases. Throughout his career, Professor Giacca has held numerous prestigious leadership roles. He has served as Professor of Molecular Biology at the Università degli Studi di Trieste, Italy, Professor of Cardiovascular Sciences at the King's College London, UK and Director-General of the International Centre for Genetic Engineering and Biotechnology (ICGEB). Among his most remarkable achievements is the identification of a crucial #microRNA promoting heart regeneration, along with the discovery of unexpected novel targets for cardiac repair (Ruozi et al, Science Translational Med 2022; Eulalio et al, Nature 2012; Lesizza et al Circ Res. 2017; Gabisonia et al, Nature. 2019).

This is an important indicator of the relevance and quality of their scientific work - and proof of the excellent research taking place at our university. 🚀🚀🚀 Fantastic team effort to discover the role of ubiquitination and selective autophagy in combatting defective splicing now published in Science: A team around Institute of Biochemistry II (IBC2) Director Ivan Đikić and Cristian Prieto-Garcia reveals how cells deploy ubiquitin-proteasome system and ER-phagy to combat the harmful effects of cryptic splicing, protecting against accumulation of misfolded proteins and preserving cellular health. #Splicing is a precisely regulated process facilitated by the #spliceosome, a large protein complex composed of more than 200 proteins. Defects in this dynamic process cause diseases such as retinitis pigmentosa (#RP), a genetic disorder that leads to progressive retina degeneration due to aberrant splicing in photoreceptor cells. 🔬The team around Đikić now discovered a novel mechanistic link between the deubiquitinase #USP39 and impaired RNA splicing. Using zebrafish and cellular models, they demonstrated that USP39 depletion mimics RP-like phenotypes with retinal degeneration. USP39 deficiency led to activation of cryptic 5’ splice sites, resulting in the production of aberrant protein isoforms that are prone to misfolding and aggregation. The misfolded proteins and protein aggregates disrupt cellular proteostasis and trigger cellular stress responses. 🔬 The team analyzed the repertoire of stress responses following USP39 depletion and found that cells ramped up #ubiquitination and #ERphagy, a specialized form of selective #autophagy targeting the endoplasmic reticulum (ER). By this, misfolded proteins can be removed and ER stress alleviated. If the proteotoxic burden cannot be successfully managed, then this leads to prolongation of ER stress, activation of the unfolded protein response (UPR), and, ultimately, apoptotic cell death. ➡ The result highlights the central role of the ubiquitin and autophagy systems in the pathology of spliceosome-associated diseases such as RP. 👏 Lingampally and colleagues from the Institute for Lung Health identified a subcluster of activated myofibroblasts (aMYFs) that may have significant implications the future management of idiopathic pulmonary fibrosis (IPF). #PaperOfTheMonth IPF is a progressive, irreversible lung disease characterized by excessive extracellular matrix deposition. From a cell biology perspective, aMYFs are thought to secrete extracellular matrix and drive structural remodeling. Using genetic lineage tracing and single-cell transcriptomics, Lingampally and colleagues investigated the origin of aMYFs and their differentiation trajectories during the process of fibrosis formation and resolution. For the first time, the researchers demonstrate the heterogeneity of aMYFs, identifying subclusters with differing roles - some exhibit distinct pro-alveologenic, while others pro-fibrotic properties. Lipofibroblasts play a major role in emergence and fate of aMYFs during fibrogenesis and resolution, respectively.
