The tools used for #prokaryotic single-cell isolation are mostly designed for aerobic conditions. Here again, we are facing challenges when working with strictly #anaerobic microorganisms like #methanogens. The Department of Microbiology and #Archaea Centre at the Universität Regensburg developed a wonderful tool to solve this issue: an anaerobic optical tweezer system. By using an inverted #microscope equipped with a Nd-YAG #laser, it is now possible to catch individual cells under anaerobic conditions and transfer them into sterile culture media. The creation of a pure culture by definition! This established, yet rare system is the only tool for single-cell isolation of certain #anaerobes from #biofilms or multi-species enrichments. It’s great that our microfluidcs team from Hahn-Schickard and the Albert-Ludwigs-Universität Freiburg Marvin Heyer, Jan Lüddecke and Fabian Lickert take part in the #microTAS conference in Canada from October 13 to 17. It is a key event for science because it brings together experts from various fields, including microfluidics, nanotechnology, chemistry, biology, and engineering. In his talk, Jan presents the “V-Disk” - a microfluidic cartridge which isolates extracellular vesicles (EVs) from 3 ml of blood or 1 ml of plasma, offering high yield and purity. It combines cation-exchange chromatography (CEX), filtration, and mixed-mode chromatography (MMC). Using waste liquid for pressure, the V-Disk increases EV yield fourfold compared to traditional methods, with similar purity, and shows potential for standardizing EV purification in clinical settings. 👉 For more information, see link in comment. #microfluidics #vesicles #biomarkers #lifesciences
